In short

Ivacaftor is a cystic fibrosis transmembrane conductance regulator potentiator that is used to treat cystic fibrosis. In the European Union, ivacaftor tablets are indicated as monotherapy for patients aged 6 years and older weighing 25 kg or more with an R117H CFTR mutation or specific gating mutations, or in combination regimens with tezacaftor/ivacaftor or ivacaftor/tezacaftor/elexacaftor for patients aged 6 years and older with specific mutations. European Union approval also includes ivacaftor granules as monotherapy for infants aged at least 1 month, toddlers, and children weighing 3 kg to less than 25 kg with an R117H CFTR mutation or specific gating mutations. In the United States, ivacaftor is indicated for the treatment of cystic fibrosis in patients aged 1 month and older who have at least one mutation in the CFTR gene that is responsive to ivacaftor potentiation based on clinical or in vitro assay data.

1 INDICATIONS AND USAGE KALYDECO is indicated for the treatment of cystic fibrosis (CF) in patients aged 1 month and older who have at least one mutation in the CFTR gene that is responsive to ivacaftor potentiation based on clinical and/or in vitro assay data [see Clinical Pharmacology (12.1) and Clinical Studies (14) ] . If the patient's genotype is unknown, an FDA-cleared CF mutation test should be used to detect the presence of a CFTR mutation followed by verification with bi-directional sequencing when recommended by the mutation test instructions for use.
