In short

In the European Union, trametinib is indicated as a monotherapy or in combination with dabrafenib to treat adults and adolescents aged 12 years and older with unresectable or metastatic melanoma with a BRAF V600 mutation, and as an adjuvant treatment for Stage III melanoma with the same mutation following complete resection. In the European Union, trametinib in combination with dabrafenib is also indicated for adults with advanced non-small cell lung cancer with a BRAF V600 mutation, and for adults with locally advanced or metastatic differentiated thyroid cancer with a BRAF V600E mutation who are refractory to or ineligible for radioactive iodine and have progressed during or after prior systemic therapy. Additionally, in the European Union, trametinib in combination with dabrafenib is indicated for paediatric patients aged 1 year and older with low-grade glioma with a BRAF V600E mutation requiring systemic therapy, or with high-grade glioma with the same mutation who have received at least one prior radiation or chemotherapy treatment. In the United States, trametinib is indicated as a single agent for BRAF-inhibitor treatment-na#ve patients with unresectable or metastatic melanoma with BRAF V600E or V600K mutations, and in combination with dabrafenib for unresectable or metastatic melanoma, adjuvant treatment of melanoma with lymph node involvement following complete resection, metastatic non-small cell lung cancer, and locally advanced or metastatic anaplastic thyroid cancer. In the United States, the combination is also approved under accelerated approval for adult and pediatric patients 1 year of age and older with unresectable or metastatic solid tumors with a BRAF V600E mutation who have progressed following prior treatment and have no satisfactory alternative treatment options.
